Glia. 2008 Sep 23; Authors: Tusell JM, Ejarque-Ortiz A, Mancera P, Solà C, Saura J, Serratosa J The cdk inhibitor p21(Cip1), also named p21(Cip1/Waf1), is intimately involved in coupling growth arrest to cellular differentiation in several cell types. p21(Cip1) is a multifunctional protein that might regulate cell-cycle progression at different levels. In a recent study, we found no differences in the rate of proliferation between glial cells from wild-type and p21(Cip1-/-) mice. In the present study, we examined differences in glial activation between glial cells from wild-type and p21(Cip1-/-) mice, using mixed glial cultures, microglia-enriched cultures, and astrocyte-enriched cultures. We compared the effect of lipopolysaccharide and two forms (oligomeric and fibrillar) of the 1-42 beta-amyloid peptide on glial activation. We observed an attenuation of nuclear translocation of the nuclear factor kappa-B in p21(Cip1-/-) glial cells, when compared with glial cells from wild-type mice. In contrast, tumor necrosis factor-alpha release was enhanced in p21(Cip1-/-)microglial cells. In addition glial activation induced by lipopolysaccharide and the fibrillar form of the 1-42 beta-amyloid peptide upregulated p21(Cip1). To determine whether hematopoietic cells or non-hematopoietic cells provides the Dll1-mediated signals to primitive hematopoietic cells, we transplanted lineage(-)c-kit(+)Sca-1(+) (KSL) bone marrow cells derived from wild-type (Dll1(+/+)) GFP-transgenic mice into lethally irradiated Dll1 conditional knockout (cKO) mice. After transplantation, we examined the kinetics of hematopoietic reconstitution and found that although the frequency of stem/progenitor subsets and of more mature lymphoid, myeloid, and erythroid lineages were normal, the donor-derived hematopoietic cells failed to differentiate into MZB cells. We further demonstrated that while the splenic stromal cells of wild-type mice expressed Dll1 molecule, the splenic stromal cells of recipient Dll1 cKO mice deleted the expression of Dll1. These results suggesting that the expression of Dll1 in splenic non-hematopoietic stromal cells, but not hematopoietic cells, is essential for the development of MZB cells. We have assessed the cellular and molecular characteristics of aged epidermal keratinocytes in monolayer cultures and in skin by measuring their morphological, fluorometric and biochemical properties. Light and electron microscopy, as well as flow cytometry, revealed increase in cell size, changes in cell shape, alterations in mitochondrial structure and cytoplasmic content. We showed that the expression of 16 biochemical markers was altered in aged cultured cells and in tissues, including caspases 1 and 3 and beta-galactosidase activities, immunoreactivities of p16, Ki67, 20S proteasome and effectors of the Fas-dependent apoptotic pathway. Aged cells diversity, and individual variability of aging markers, call for a multifunctional assessment of the aging phenomenon, and of its modulation by drugs. As a test case, we have measured the effects of Dead Sea minerals on keratinocyte cultures and human skin, and found that they stimulate proliferation and mitochondrial activity, decrease the expression of some aging markers, and limit apoptotic damage after UVB irradiation. We assessed whether bright polychromatic lamps enriched in the short-wavelength portion of the visible light spectrum could produce larger phase advances than standard bright white lamps. METHODS: Twenty-two healthy young adults received either a bright white or bright blue-enriched 2-h phase advancing light pulse upon awakening on each of four treatment days. On the first treatment day the light pulse began 8h after the dim light melatonin onset (DLMO), on average about 2h before baseline wake time. On each subsequent day, light treatment began 1h earlier than the previous day, and the sleep schedule was also advanced. RESULTS: Phase advances of the DLMO for the blue-enriched (92+/-78min, n=12) and white groups (76+/-45min, n=10) were not significantly different. CONCLUSION: Bright blue-enriched polychromatic light is no more effective than standard bright light therapy for phase advancing circadian rhythms at commonly used therapeutic light levels. Hurricane Ike threatened to wreak havoc with international commerce along the Gulf Coast, as the port of Houston - the nation's second busiest - prepared Thursday to shut down cargo operations through the weekend, and airlines canceled flights to the region. The Port of Houston Authority said it relied on information from the Coast Guard in planning the shutdown. The 25-mile-long Houston port receives consumer goods - "anything that you could buy at Wal-Mart or Target," said port spokeswoman Lisa Whitlock - plus automobiles, industrial equipment and other cargo from around the world and ships out vast amounts of petrochemicals and agricultural products. Officials told cargo operators to prepare their vessels to leave the port if requested by the Coast Guard. "It's safer for vessels to go out to sea to get away from turbulent waters," Whitlock said. "If they're stuck in port, they have nowhere to go and can bump against docks." The port is expected to reopen Monday. The approaching storm caused gasoline prices to surge on concerns that the storm could damage Texas refineries, or at the least halt production for several days. As an aside in his observations on Tuesday’s iPod event , our good friend John Gruber over at Daring Fireball snuck in a remark about an impending Apple laptop update: Those of you holding out for a new lineup of MacBooks will have to wait until October 14, according to sources who, as they say, are familiar with Apple’s hardware plans. What’s this? Rumors from the Gruber? Well, John’s not the kind of guy to post spurious facts willy-nilly: notice that he slipped it into a post on another subject instead of putting it under a screaming big Helvetica headline. Since then, a bunch of other sites have gone all atwitter for ferreting out this cleverly hidden detail . For what it’s worth, our sources confirm that Gruber’s right on the money, and even if they didn’t , such a prediction is hardly out of left field. Apple revved MacBooks in late October/early November in both 2006 and 2007 , and the last speed bumps the MacBook line saw was back in February , suggesting they’re due for an update—especially with Intel having recently pimped new mobile chips .
