J Immunotoxicol. 2008 Jul;5(3):315-35 Authors: Sheets RL, Stein J, Bailer RT, Koup RA, Andrews C, Nason M, He B, Koo E, Trotter H, Duffy C, Manetz TS, Gomez P The Vaccine Research Center has developed vaccine candidates for different diseases/infectious agents (including HIV-1, Ebola, and Marburg viruses) built on an adenovirus vector platform, based on adenovirus type 5 or 35. To support clinical development of each vaccine candidate, pre-clinical studies were performed in rabbits to determine where in the body they biodistribute and how rapidly they clear, and to screen for potential toxicities (intrinsic and immunotoxicities). The vaccines biodistribute only to spleen, liver (Ad5 only), and/or iliac lymph node (Ad35 only) and otherwise remain in the site of injection muscle and overlying subcutis. Though approximately 10(11) viral particles were inoculated, already by Day 9, all but 10(3) to 10(5) genome copies per mu g of DNA had cleared from the injection site muscle. By three months, the adenovector was cleared with, at most, a few animals retaining a small number of copies in the injection site, spleen (Ad5), or iliac lymph node (Ad35). This pattern of limi ... Transplant Proc. 2008 Jul-Aug;40(6):1965-71 Authors: Di Benedetto F, Di Sandro S, De Ruvo N, Berretta M, Montalti R, Guerrini GP, Ballarin R, De Blasiis MG, Spaggiari M, Smerieri N, Iemmolo RM, Guaraldi G, Gerunda GE INTRODUCTION: Highly active antiretroviral therapy (HAART) has been able to improve the immune system function and survival of HIV patients with a consequent increase in the number of HIV patients affected by end-stage liver disease (ESLD). Between June 2003 and October 2006, 10 HIV-positive patients underwent liver transplantations in our Center. METHODS: All patients were treated with HAART before transplantation; treatment was interrupted on transplantation day and was restarted once the patients' conditions stabilized. Five patients were hepatitis C Virus (HCV)-positive, 3 were hepatitis B Virus (HBV)-positive, and 2 were HBV-HCV coinfected. HIV viral load before transplantation was <50 copies/mL in all cases. CD4+ cell count before transplantation ranged between 144 and 530 c/microL. Immunosuppression was based on Cyclosporine (CyA) and steroid weaning for 8 patients, and on Tacrolimus and steroid weaning for 2 patients. RESULTS: Five patients were cytomegalovirus (CMV)-positive pp65 antigenemia posttransplantation, and 1 patient was EBV-positive; 2 patients had a coinfection with HHV6. Four patients suffered from a cholestati ... Source: www.ncbi.nlm.nih.gov --- 5 days ago
Related Articles What is the biopsychosocial role of human immunodeficiency Virus positivity in patients with end-stage liver disease who undergo orthotopic liver transplantation? Transplant Proc. 2008 Jul-Aug;40(6):1910-2 Authors: Barbanti Silva V, Ferrara M, Mazzi F, Pingani L, Cocchi S, Romano A, Masetti M, Gerunda GE, Guaraldi G, Rigatelli M INTRODUCTION: Since 2003 the National Research Program for Solid Organ Transplantation in patients with human immunodeficiency Virus (HIV) is active at our liver transplantation Center. Patients with HIV who enter this protocol are assessed by the Consultation Liaison Psychiatry Service. The aim of the present study was to evaluate their psychiatric comorbidity. METHODS: An observational prospective study was conducted comparing end-stage liver disease (ESLD) patients with and without HIV. After the assessment, the psychiatrist compiled the Transplant Evaluation Rating Scale (TERS) and the Montgomery Asberg Depression Rating Scale (MADRS). Baseline evaluation was made before inclusion on the OLT waiting list and the follow-up evaluation was made 12 months later. RESULTS: From January 2003 to December 2006 we assessed 553 patients: 39 (6%) with HIV and 361 (94%) without HIV. The 2 groups were homogeneous for gender (75% of male patients; P=not significant [NS]) but not for age (46+/-5 vs 56+/-9; P=NS). Psychiatric history was negative in 176 (49%) patients without HIV and in 6 (15%) patients wi ... Source: www.moreover.com --- 33 days ago

The awarding of the Nobel Prize in medicine on Monday writes a postscript to a bitter scientific dispute in the 1980s over who deserved credit for discovering HIV and the resulting test to screen blood for it. Among Monday's three honorees is Dr. Luc Montagnier (Center), who with fellow Frenchman Francoise Barre-Sinoussi was cited for discovery of the AIDS Virus. (Getty-AFP photo / Issouf Sanogo) ...
