Mol Cell Biol. 2008 Oct 13; Authors: Wurster AL, Pazin MJ During T helper cell differentiation, distinct programs of gene expression play a key role in defining the immune response to an environmental challenge. How chromatin Remodeling events at the associated cytokine loci control differentiation is not known. We found that the ATP-dependent Remodeling enzyme subunit BRG1 was required for T helper 2 (Th2) differentiation and Th2 cytokine transcription. BRG1 binding to cytokine genes was regulated by the extent of differentiation, the extent of activation and cell fate. BRG1 was required for some features of the chromatin structure in target genes (DNase I hypersensitivity and histone acetylation), suggesting that BRG1 Remodeling activity was directly responsible for changes in gene expression. NFAT and STAT6 activity were required for BRG1 recruitment to the Th2 LCR, and STAT6 associated with BRG1 in a differentiation-inducible manner, suggesting direct recruitment of BRG1 to the bound loci. Together, these findings suggest BRG1 interprets differentiation signals and plays a causal role in gene regulation, chromatin structure, and cell fate. Here, we describe a protein encoded by a novel germ-cell-specific gene, Mm.290718/ZFP541, identified from the mouse spermatocyte UniGene library. The protein contains specific motifs and domains potentially involved in DNA binding and chromatin reorganization. An antibody against Mm.290718/ZFP541 revealed the existence of the protein in testicular spermatogenic cells (159 kDa), but not testicular and mature sperm. Immunostaining analysis of cells at various stages of spermatogenesis consistently showed that the protein is present in spermatocytes and round spermatids only. Transfection assays and immunofluorescence studies indicate that the protein is localized specifically in the nucleus. Proteomic analyses performed to explore the functional characteristics of Mm.290718/ZFP541 showed that the protein forms a unique complex. Other major components of the complex included histone deacetylase 1 (HDAC1) and heat-shock protein A2. We used two-dimensional liquid chromatography fractionation, followed by protein identification by nanoLC-MS/MS. This analysis revealed distinct changes in proteins implicated in neurotransmission and neurogenesis, in cytoskeleton dynamics and in the regulation of gene expression and chromatin Remodeling. Loss of bone density later may be associated with stress shielding or normal bone loss of aging. We evaluated Remodeling changes over time with a proximally hydroxyapatite-coated tapered titanium stem. We evaluated plain radiographs of 143 hips for cancellous condensation, cortical hypertrophy, cortical porosis, cortical index, and canal fill at early postoperative, 5, 10, and 15 years. Average age was 51 years at THA; 69 patients (77 hips) (53%) were women; and 102 hips (71%) had primary osteoarthrosis. Based on radiographic findings at 15 years, hips were divided into three subgroups: 43 (30%) demonstrated minimal Remodeling changes; 53 (37%) demonstrated cortical hypertrophy evident before 5 years; and 47 (33%) demonstrated additional late Remodeling and cortical porosis, most often after 10 years. Hips with poorer bone (Dorr Types B or C) and, when including only hips with osteoarthrosis, more female hips had cortical porosis at 15 years. To further explore new therapeutic strategies to improve learning and memory, we hypothesized that a potential mechanism to reinstate learning and memory in a degenerated brain would be to up-regulate the plasticity and function of the remaining neurons. Environmental enrichment (EE) was previously shown to facilitate memory formation and brain plasticity in wild-type rodents. To determine the effect of EE on learning behavior after neuronal loss had already occurred, we induced p25 in 11-month-old CK-p25 Tg mice for 6 weeks, followed by EE for 4 weeks. Despite a comparable extent of brain atrophy, EE-treated CK-p25 Tg mice showed marked increased associative and spatial learning when compared to the nonenriched CK-p25 Tg mice. Enhanced learning behavior was associated with increased dendritic branching and synapse number. Although these results suggested that learning behavior could be improved by EE, we further investigated whether EE is beneficial for retrieving long-term memories. We applied a single contextual fear-conditioning training, which was previously shown to result in a stable long-term memory to 11-month-old, uninduced CK-p25 Tg mice. The mice were kept in the home cage for 4 weeks to allow the consolidation of hippocampus-independent long-term memories.
