Related Articles Identification of melatonin-regulated genes in the ovine pituitary Pars tuberalis, a target site for seasonal hormone control. Endocrinology. 2008 Jul 31; Authors: Dupré SM, Burt DW, Talbot R, Downing A, Mouzaki D, Waddington D, Malpaux B, Davis JR, Lincoln GA, Loudon AS The Pars tuberalis (PT) of the pituitary gland expresses a high density of melatonin (MEL) receptors and is believed to regulate seasonal physiology by decoding changes in nocturnal melatonin secretion. Circadian clock genes are known to be expressed in the PT in response to the decline (Per1) and onset (Cry1) of MEL secretion, but to date little is known of other molecular changes in this key MEL target site. To identify transcriptional pathways that may be involved in the diurnal and photoperiod-transduction mechanism, we performed a whole genome transcriptome analysis using PT RNA isolated from sheep culled at three time points over the 24h cycle under either long or short photoperiods. Our results reveal 153 transcripts where expression differs between photoperiods at the light/dark transition and 54 transcripts where expression level was more globally altered by photoperiod (all time points combined). Cry1 induction at night was associated with up-regulation of genes coding for NeuroD1 (Neurogenic differentiation factor 1), Pbef/Nampt, (nicotinamide phosphoribosyltransferase), Hif1alpha (hypoxia-inducible factor-1alpha) and Kcnq5 (K(+) channel) an ...

Prostatic trypsin-like kallikrein-related peptidases (KLKs) and other prostate-expressed tryptic proteinases as regulators of signalling via proteinase-activated receptors (Pars). Biol Chem. 2008 Jun 20; Authors: Ramsay AJ, Reid JC, Adams MN, Samaratunga H, Dong Y, Clements JA, Hooper JD Abstract The prostate is a site of high expression of serine proteinases including members of the kallikrein-related peptidase (KLK) family as well as other secreted and membrane-anchored serine proteinases. It has been known for some time that members of this enzyme family elicit cellular responses by acting directly on cells. More recently it has been recognised that for serine proteinases with specificity for cleavage after arginine and lysine residues (trypsin-like or tryptic enzymes) these cellular responses are often mediated by cleavage of members of the proteinase-activated receptor (PAR) family - a four member sub-family of G protein-coupled receptors. Here we review the expression of Pars in prostate, the ability of prostatic trypsin-like KLKs and other prostate-expressed tryptic enzymes to cleave Pars as well as the prostate cancer-associated consequences of PAR activation. In addition, we explore the dysregulation of trypsin-like serine proteinase activity through the loss of normal inhibitory mechanisms and potential interactions between these dysregulated enzymes leading to aberrant PAR activation, intracellular signalling and cancer prom ...

Kallikreins and proteinase-mediated signaling: proteinase-activated receptors (Pars) and the pathophysiology of inflammatory diseases and cancer. Biol Chem. 2008 Jun 20; Authors: Hollenberg MD, Oikonomopoulou K, Hansen KK, Saifeddine M, Ramachandran R, Diamandis EP Abstract Proteinases like thrombin and trypsin can affect tissues by activating a novel family of G-protein-coupled Proteinase-Activated Receptors (Pars 1 to 4) by exposing a 'tethered' receptor-triggering ligand (TL). Work with synthetic TL-derived PAR peptide sequences (PAR-APs) that stimulate Pars 1, 2 and 4 has shown that PAR activation can play a role in many tissues, including the gastrointestinal tract, kidney, muscle, nerve, lungs and the central and peripheral nervous systems and can promote tumor growth and invasion. Pars may play roles in many settings including cancer, arthritis, asthma, inflammatory bowel disease, neurodegeneration and cardiovascular disease, as well as in pathogen-induced inflammation. In addition to activating or dis-arming Pars, proteinases can also cause hormone-like effects by PAR-independent mechanisms, like activation of the insulin receptor. In addition to proteinases of the coagulation cascade, recent data suggest that members of the family of kallikrein-related peptidases (KLK) represent endogenous PAR regulators. In summary: (1) proteinases are like hormones, signaling in a paracrine and endocrine manner via Pars or other mechanisms, ...

BETHESDA, Md. — Tom Pernice Jr. was easy to ignore Saturday in the AT&T National until he wound up in the lead. All he did was make 15 Pars on an explosive afternoon of birdies and bogeys at Congressional, and his lone complaint was that he made only two putts longer than his shadow. One was from 4½ feet for par, another from 7 feet for birdie. But it was that steady hand that gave him a 1-under 69 and a one-shot lead over Steve Stricker, leaving the 48-year-old Pernice optimistic about winning for the first time in seven years. ”It will be interesting to see how it goes tomorrow,“ Pernice said. ”But I'm looking forward to it.“ ...
