The DNA damage response and DNA repair pathways are critical pillars of organismal defenses, minimizing somatic mutations in both post-mitotic and mitotic cells. These genomic maintenance systems not only prevent the premature emergence of cancers but may also maintain normal tissue function and organismal Longevity. Genetic defects in a number of DNA repair and DNA damage response genes often leads to a dramatic increase in cancer incidence; in other cases, premature aging or progeroid syndromes may be induced. In this review, we discuss two recent reports of two nucleotide excision repair-deficient models that exhibit dramatic premature aging and shortened Longevity. The DNA repair defects were also associated with a significant inhibition of the growth hormone/insulin-like growth factor 1 (GH/IGF-1) axis, an endocrine signaling pathway shown to influence aging and Longevity in both vertebrates and invertebrates. Potential mechanisms of how DNA damage might affect IGF-1 signaling and aging are discussed, with a particular emphasis on the role of such signaling alterations in the adult tissue stem cell compartments. Two new studies have found the molecular link between circadian rhythm and metabolism -- and it turns out to be a protein targeted by resveratrol, a potentially Longevity-enhancing drug. Neither of the studies, published yesterday in Cell, tested the metabolic effects of resveratrol, but they strike a note of caution for those -- including myself -- who expect resveratrol to be the first of an entire class of drugs that slow the physical and mental deterioration of aging. Resveratrol's target is SIRT1, one of a group of proteins called sirtuins that maintain cellular function. But the researchers -- one group from the University of California, Irvine and another from the University of Geneva -- say that SIRT1 is also an important piece of our biological clocks: it links cell-level ciradian rhythms with body-level physiology, keeping appetite and wakefulness in a smoothly-regulated cycle. Whether sirtuin-activating drugs will interfere with this is unknown, but it shouldn't be hard to test. (Talk about a side effect: you'll live long and healthily, but have trouble sleeping and get hungry at inconvenient times! I'd take it.) (NaturalNews) Sticking more closely to a Mediterranean diet may lead to a longer life, according to a new study published in the Archives of Internal Medicine . The researchers say that the study is the first to examine a connection between the diet and mortality in a U.S. population. Researchers examined data on more than 160,000 women and 200,000 men who had filled out detailed questionnaires about their eating habits, weight, age, smoking status and other factors known to affect health. The researchers then ranked participants based on how closely they held to either a traditional Mediterranean diet or an alternate Mediterranean diet. Participants scored higher for adherence to a traditional Mediterranean diet if they ate more fruits and nuts, grains, beans, fish and vegetables (excluding potatoes), and if they ate less dairy, meat or saturated fat. Those who drank a moderate amount of alcohol, defined as five to 25 grams per day for women and 10 to 50 grams per day for men, also scored higher. The alternative Mediterranean diet was scored similarly, but included only whole (not refined) grains, did not include dairy products, counted fruits and nuts separately, used the same alcohol standard for men and women, and deducted points for meat only if it was red or processed.
