The deduced KIAA0319 protein contains several polycystic kidney disease (PKD) domains which may mediate the interaction between neurons and glial fibres during neuronal migration. We have previously reported the presence of several alternative splicing variants, some of which are predicted to affect the deduced protein. In this study, we over-expressed constructs containing the main form (A) and two alternative variants (B and C) of KIAA0319. We show that the full-length KIAA0319 (A) is a type I plasma membrane protein, a topology consistent with its proposed function in neuronal migration. The oligomeric status of KIAA0319 is mainly dimeric, and this condition depends on the cysteine-rich regions of the protein, especially the transmembrane (TM) domain and surrounding sequence. KIAA0319 is highly glycosylated in different mammalian cell lines. The central region including the PKD domains is N-glycosylated. Only few data are available on the pathomechanisms of this specific dysfunction, assuming - among others - that Dyslexia might be a disconnection syndrome of anterior and posterior brain regions involved in phonological and orthographic aspects of the reading process, as well as in the integration of phonemes and graphemes. Therefore, diffusion tensor imaging (DTI) and voxel-based morphometry (VBM) were used to verify the hypothesis of altered white and gray matter structure in German dyslexic adults. DTI revealed decreased fractional anisotropy (FA) in bilateral fronto-temporal and left temporo-parietal white matter regions (inferior and superior longitudinal fasciculus). Significant correlations between white matter anisotropy and speed of pseudoword reading were found. In dyslexics, gray matter volumes (as measured by VBM) were reduced in the superior temporal gyrus of both hemispheres. Researchers suggest that taking part in specially developed computer-aided exercises can help prevent the onset of Dyslexia. In a study conducted at the Centre of Excellence in Learning and Motivation Research, it was found that children with parents who have learning difficulties are more likely to develop Dyslexia themselves. Lead researcher Professor Heikki Lyytinen of the University of Jyvaskyla recommends that children who have a perceived risk of developing learning difficulties, or those who had already shown signs of this should make use of the exercises, developed by the research centre, as early as possible. 'The best time to start these exercises is the latter part of the pre-school age, but it's not too late even after the children have started school,' he stated.
