Int J Pharm. 2008 Sep 9; Authors: Kumar V, Sharma VK, Kalonia DS Feasibility studies were performed to develop a process for obtaining stable Dry protein formulations based on in situ polyethylene glycol (PEG)-induced precipitation and Vacuum drying of interferon alpha-2a (IFNalpha2a) solution in a vial. Using a laboratory scale freeze dryer, the process was carried out in two phases: first, protein solution containing PEG was concentrated to achieve protein precipitation, and second, remaining water was removed by further reducing the chamber pressure. Drying conditions, i.e. temperature and pressure, and solution composition were selected to ensure maximal precipitation (solubility of IFNalpha2a), to achieve precipitation without boiling, and to ensure stability. Dried formulations were subjected to stability studies (40 degrees C). Concentration and precipitation could be achieved at a fast rate by utilizing pressures slightly above the vapor pressure of water. Fluorescence and circular dichroism (CD) studies showed that precipitated IFNalpha2a maintained its native structure. Fourier transform infrared spectroscopy (FTIR) studies showed that IFNalpha2a when dried in the presence of trehalose, maintained its secondary structure. We report photoinitiated chemical vapor deposition (piCVD), a gentle synthetic method for the preparation of ultra thin films (~100 nm) of the hydrogel poly(hydroxyethyl methacrylate) (pHEMA). piCVD occurs near room temperature and requires only mild Vacuum conditions. The deposited films swell rapidly and reversibly in buffer solution and the swelling properties can be controlled via the deposition conditions. Analysis of the swelling data indicates that the mesh size of the hydrogel creates a selectively permeable coating. The mesh is large enough to allow small molecule analytes to permeate the film but small enough to prevent the transport of large biomolecules such as proteins. X-ray photoelectron spectroscopy (XPS) shows that the films decrease nonspecific adhesion of the protein albumin by nearly eight-fold over bare silicon. A Dry process, piCVD is suitable for coating particles with diameters as small as 5 m. Ooh, that smell First things first: Get the water and mud out with a wet-Dry Vacuum or broom. Then spray all surfaces with lukewarm water, starting with the floor and working up. Scrub the surfaces with hot water and a heavy-duty cleaner, then rinse using a brush moistened with a solution of two tablespoons of chlorine bleach per gallon of water. Repeat scrubbing and rinsing until the odor is gone. Disinfecting duties If there is sewage in the water (or you just want to take precautions), clean with a solution of one part chlorine bleach to nine parts water. Use rock salt (one cup per gallon of water) on plaster and painted drywall, but not on concrete floors, which the salt will damage. Rug redemption If a carpet has been soaked with water and sewage, ditch it. Wear rubber gloves and be careful not to scrape your skin, to avoid infection. For slight water damage, remove the carpet if possible, roll it up, and take it outside. A fire that tore through a famous 19th century British ship was likely caused by a Vacuum cleaner used for renovation work bursting into flames, police said Tuesday. Investigators found no evidence of arson for last year's fire that threatened to destroy the Cutty Sark, a historic three-masted tea clipper that sits in Dry dock on the banks of London's River Thames. The most likely causes were a Vacuum cleaner catching fire after it was accidentally left on over the weekend or electrical equipment that overheated, according to a report released Tuesday. "It is the view of the inquiry that the cause of the fire was accidental," police inspector Dave Garwood told reporters. Firefighters battled the blaze for more than two hours early on May 21, 2007, near Britain's National Maritime Museum at Greenwich. The inferno sent clouds of black smoke over the leafy southeast London borough, but much of the Cutty Sark's historic material - including its masts, deckhouses, and most of the ship's hull planking - had already been sent to the Chatham Dockyards, east of London, for conservation work. The trust said less than 2 percent of the vessel's original material was lost to the flames. "It could have been much, much worse," the Cutty Sark Trust's chief executive Richard Doughty said.
