We present the nuclear magnetic resonance (NMR) solution structure of recombinant human VDAC-1 reconstituted in Detergent micelles. It forms a 19-stranded beta barrel with the first and last strand parallel. The hydrophobic outside perimeter of the barrel is covered by Detergent molecules in a beltlike fashion. In the presence of cholesterol, recombinant VDAC-1 can form voltage-gated channels in phospholipid bilayers similar to those of the native protein. NMR measurements revealed the binding sites of VDAC-1 for the Bcl-2 protein Bcl-x(L), for reduced beta-nicotinamide adenine dinucleotide, and for cholesterol. MATERIALS AND METHODS: Nine phototoxic and one non-phototoxic compounds (200 microM), dissolved in Tween 20, sodium laurate, or sodium dodecyl sulfate (SDS) micellar solution, were exposed to UVA/B light (250 W/m2), and singlet oxygen generation was monitored by RNO bleaching methodology. Photochemical properties of photosensitizers were also evaluated by UV measurement, and the interaction of photosensitizers with surfactant micelles was assessed by Z-potential and NMR spectroscopic analyses. RESULTS: All phototoxic compounds tended to generate singlet oxygen under light exposure in the all micellar solutions tested. Heart diseases resulting in heart failure are among the leading causes of morbidity and mortality in the Western world and can result from either systemic disease (e.g., hypertensive heart disease, ischemic heart disease) or specific heart muscle disease (e.g., dilated cardiomyopathy/DCM). Subproteome analysis of such disease subsets affords a reduction in sample complexity, potentially revealing biomarkers of cardiac failure that would otherwise remain undiscovered in proteome wide studies. Label-free nanoscale LC-MS has been applied in this study to validate a Triton X-114-based phase enrichment method for cardiac membrane proteins. Annotation of the subcellular location combined with GRAVY score analysis indicates a clear separation between soluble and membrane-bound proteins with an enrichment of over 62% for this protein subset. LC-MS allowed confident identification and annotation of hydrophobic proteins in this control sample pilot study and demonstrates the power of the proposed technique to extract integral membrane-bound proteins. I remember a few years back thinking it so odd to see commercials for products like Snickers and whatnot that touted how the products were the official candy bars and whatevers of the U.S. Olympic team. Really, U.S. Olympiads need an official candy bar? So imagine how surprised I was to learn that this year, the U.S. Olympic team has an official laundry Detergent. It's Win High-Performance Sport Detergent . The makers say it is specially designed to get the sweat and stink out of athletic gear that features moisture-wicking fabrics, including Nike Dri-Fit, Under Armour, Coolmax and more. I'm not so sure that I need to use a special Detergent for my fitness gear.
