J Immunol. 2008 Oct 15;181(8):5199-203 Authors: Rudd BD, Brien JD, Davenport MP, Nikolich-Zugich J TLR ligands are among the key stimuli driving the optimal dendritic cell (DC) maturation critical for strong and efficacious T cell priming. In this study, we show that part of this effect occurs via increased TCR triggering. Pretreatment of DCs with TLR ligands resulted in the triggering of many more TCRs in responding CD8(+) T cells. Importantly, even when DCs expressed the same amount of cognate peptide-MHC (pMHC) molecules, TLR ligand treatment resulted in down-regulation of larger numbers of TCR molecules. This was independent of the up-regulation of costimulatory, adhesion or cytokine molecules or the amount of noncognate pMHCs. Rather, DCs pretreated with TLR ligands exhibited increased stability of cognate pMHCs, enabling extended TCR triggering. These findings are of potential importance to T cell vaccination. The effect of experimental parameters on the signal Decay in double-PGSE experiments: Negative diffractions and enhancement of structural information. J Magn Reson. 2008 Sep 17; Authors: Shemesh N, Cohen Y Double pulsed gradient spin echo (d-PGSE) experiment has been recently suggested for detecting microscopic anisotropy in macroscopically isotropic samples. This sequence is complex and has many variables, including, intra alia, combinations of directions and amplitudes of the pulsed gradients, diffusion times in each of the encoding periods and the mixing time period. The effect of these experimental parameters of the d-PGSE sequence was studied in an array of water filled microcapillaries of micron diameters. We found that negative diffractions occur, as indeed predicted by recently published simulations. We also found differential effects of prolongation of the mixing time between collinear and orthogonal d-PGSE experiments. The d-PGSE experiment in the collinear direction perpendicular to the long axis of the cylinder exhibited a marked dependence on the mixing time, while the orthogonal d-PGSE experiment exhibited no such dependence at all. Interestingly, one of the most important predictions by the simulations was that the d-PGSE sequence could potentially discriminate between compartments of different sizes better than the single PGSE (s-PGSE) and it seems that our experimental results indeed corroborate these predictions. Mitochondrial Decay in the Brains of Old Rats: Ameliorating Effect of Alpha-Lipoic Acid and Acetyl-L: -carnitine. Neurochem Res. 2008 Oct 10; Authors: Long J, Gao F, Tong L, Cotman CW, Ames BN, Liu J To investigate the mitochondrial Decay and oxidative damage resulting from aging, the activities/kinetics of the mitochondrial complexes were examined in the brains of young and old rats as well as in old rats fed R-alpha-lipoic acid plus acetyl-L: -carnitine (LA/ALC). The brain mitochondria of old rats, compared with young rats, had significantly decreased endogenous antioxidants and superoxide dismutase activity; more oxidative damage to lipids and proteins; and decreased activities of complex I, IV and V. Complex I showed a decrease in binding affinity (increase in K(m)) for substrates. Feeding LA/ALC to old rats partially restored age-associated mitochondrial dysfunction to the levels of the young rats. These results indicate that oxidative mitochondrial Decay plays an important role in brain aging and that a combination of nutrients targeting mitochondria, such as LA/ALC, could ameliorate mitochondrial Decay through preventing mitochondrial oxidative damage.
