New research by British and US scientists has seen repair proteins appear to jump between DNA molecules and sliding across strands to fix mistakes, like ‘jumping fleas’. Boffins at the University of Essex in the UK as well as Pittsburgh and Vermont in the US discovered this tagging proteins with quantum dots, reporting the findings in Molecular Cell . The aim of the research was to discover how a repair system worked rapidly to maintain the integrity of DNA code sequences, which were constantly being bombarded with environmental toxins. Bennett Van Houten, professor of pharmacology and chemical biology at the University of Pittsburgh, said that the way the repair system worked was an important unanswered question in the field. He said: “It has to be able to identify very small mistakes in a three-dimensional morass of gene strands. It is akin to spotting potholes on every street all over the country and getting them fixed before the next rush hour.” Two repair proteins (UvrA and UvrB) were tagged with quantum dots, which are semiconductor nanocrystals that light up in different colours. The clumped DNA was also stretched into ‘tightropes’ to see the process clearly. UvrA proteins jumped from one DNA molecule to the next, staying for about seven seconds before going on to the next one. But when a UvrA molecule formed a complex with two UvrB molecules (UvrAB), this searched more efficiently by sliding along the DNA for as long as 40 s ...

Related Articles Regulation of Inter- and Intramolecular Interaction of RNA, DNA, and Proteins by MLE. Methods Mol Biol. 2010;587:303-26 Authors: Oh H, Parrott AM, Park Y, Lee CG Drosophila maleless (MLE) is a member of helicase superfamily 2 and functions as a dosage compensation factor essential for the development of male flies. This function provides a good opportunity to investigate diverse biochemical activities associated with MLE in the context of a defined in vivo pathway, i.e., the transcriptional activation of X-linked genes. We have shown for the first time that MLE catalyzes the unwinding of both DNA and RNA and that MLE helicase activity is essential for its in vivo function. Also, we have provided evidence that MLE stimulates the transcriptional activity of roX2 on the X chromosome. We have also found that MLE interacts with dsDNA, topoisomerase II, and nucleosome. This observation supports a current model of dosage compensation: transcriptional activation of X-linked genes is causally associated with conformational change in the male X chromosome, subsequent to the targeted association of the dosage compensation complex (DCC). On its face, the establishment of a national DNA database sounds rather terrifying and Orwellian. We can’t imagine that the majority of Americans, privacy obsessed as we are, would willingly consent to such an idea — and gleefully skip on down to the DMV to give a cheek swab. Nevertheless, it appears to be an idea on the ascent. According to an opinion piece in Monday’s NYT by Yale Law student Michael Seringhaus, President Obama, in an appearance on “America’s Most Wanted,” appeared to agree that, in Seringhaus’s words, “there should be a national DNA database with profiles of every person arrested, whether convicted or not.” Seringhaus largely agrees. His argument is mostly based on the premise that coughing up a DNA sample is no more invasive to our privacy than a simple fingerprint. The sample provides no information other than, well, a unique identifier, like a PIN code. At the same, time, opines Seringhaus, the potential benefit to law-enforcement is significant: DNA samples collected from crime scenes are compared against a standing database of profiles, and matches are investigated. Obviously, the more individuals profiled in the database, the more likely a crime-scene sample can be identified, hence the president’s enthusiasm to expand the nationwide repository. Several states reportedly have taken to a technique called “familial DNA search,” which enables police to search for partial DNA matches and often provides them valu ...

By Cleo Patek Robinson To some groups of people, like Native Americans, DNA testing is used not only to establish links to ancestors but also to determine legal rights and privileges. Tribal sovereignty and legal rights are hot topics of discussion in American political arenas and are disputed in the courts all the time. States, industries and land owners themselves challenge the tribes' rights to govern their own lands and people, or to even exist at all. Most citizens do not even realize that tribes are political entities unto themselves and not just quaint little ethnic groups. And because of the legal protections afforded Native Americans, DNA testing often becomes necessary for them to be able to prove their ancestry and enjoy the rights and privileges that their tribes have earned. Because federal laws and tribal laws focus on the tribes relating as a group, their cultural continuity and their tribal land-base, many people who have Native American biological ancestors are unfortunately ineligible for federally recognized tribal status because they either don't live with the tribe, do not function with the tribe or their heritage has become mixed. DNA testing is usually the sole recourse these Native Americans have if they want to be legally recognized as a tribal member. Without this recognition, they will lose access to land, financial aid and even gaming business licenses. Some people even recommend that DNA testing become a ...

By David Kravets Josh Gerstein over at Politico sent Threat Level his piece underscoring once again President Barack Obama is not the civil-liberties knight in shining armor many were expecting. Gerstein posts a televised interview of Obama and John Walsh of America’s Most Wanted. The nation’s chief executive extols the virtues of mandatory DNA testing of Americans upon arrest, even absent charges or a conviction. Obama said, “It’s the right thing to do” to “tighten the grip around folks” who commit crime. When it comes to civil liberties, the Obama administration has come under fire for often mirroring his predecessor’s practices surrounding state secrets , the Patriot Act and domestic spying . There’s also Gitmo , Jay Bybee and John Yoo . Now there’s DNA sampling. Obama told Walsh he supported the federal government, as well as the 18 states that have varying laws requiring compulsory DNA sampling of individuals upon an arrest for crimes ranging from misdemeanors to felonies. The data is lodged in state and federal databases, and has fostered as many as 200 arrests nationwide, Walsh said. The American Civil Liberties Union claims DNA sampling is different from mandatory, upon-arrest fingerprinting that has been standard practice in the United States for decades. A fingerprint, the group says, reveals nothing more than a person’s identity. But much can be learned from a DNA sample, which codes a person’s family ties, some health ris ...

"Its just a charter of negative liberties." Obama, the constitutional-illiterate pretender-in-chief, approves of DNA testing of Americans upon arrest. What happened to innocent until proven guilty? What happened to the 4th amendment that protects Americans from unreasonable search and seizures? You would think a “constitutional law professor” would know the U.S. Constitution front to back. Obama believes anyone arrested should be forced to give up their DNA absent of charges or convictions. This includes misdemeanor charges as well. This is unconstitutional and a slippery slope situation. Politico reports: President Barack Obama’s embrace of a national database to store the DNA of people arrested but not necessarily convicted of a crime is heartening to backers of the policy but disappointing to criminal-justice reformers, who view it as an invasion of privacy. Others also worry the practice would adversely affect minorities. In an interview aired Saturday on “America’s Most Wanted,” Obama expressed strong agreement as host John Walsh extolled the virtues of collecting DNA at the time of an arrest and putting it into a single, national database. “We have 18 states who are taking DNA upon arrest,” Walsh said. “It’s no different than fingerprinting or a booking photo. … Since those states have been doing it, it has cleared 200 people that are innocent from jail.” “It’s the right thing to do,” Obama replied. “This is where the national ...

------------------------------------------------------------------------------- BACKGROUND: Seventeen years ago, Taylor was convicted of the September, 1991 murder of Raleigh prostitute Jacquetta Thomas, 26, whose body was found dumped on South Blount Street in Raleigh. Taylor, 47, said he spent the night of September 25, 1991 drinking and doing drugs with friends while he drove around southeast Raleigh to buy crack cocaine. Taylor said he believed police latched on to him for the murder because he and a friend drove along a dirt path off the same cul-de-sac where Thomas's body was found. Taylor and the friend smoked crack, but his SUV got stuck as they tried to drive away. They abandoned the SUV and walked to a nearby street to get a ride. Taylor testified they saw what they thought was a body but didn't report it to police. When Taylor returned in the morning to get the SUV, the police were already there. During several days of testimony, a parade of witnesses poked holes in the original evidence against Taylor. A SBI agent testified that while initial tests on some items from Taylor's sport utility vehicle were positive for blood, follow-up tests were negative. Those negative tests were not revealed to the jury that convicted Taylor. A dog training expert testified that the bloodhound that investigators said found the scent of the victim on Taylor's SUV was not trained in scent identification. A jailhouse snitch who said that Tayl ...
