Related Articles Cloning and expression analysis of two alternative splicing toll-like receptor 9 isoforms A and B in large yellow croaker, Pseudosciaena crocea. Fish Shellfish Immunol. 2008 Jul 19; Authors: Yao CL, Kong P, Wang ZY, Ji PF, Cai MY, Liu XD, Han XZ Toll-like receptors (TLRs) are the archetypal pattern-recognition receptors in sensing foreign pathogens. In this report, two alternative splicing isoforms of TLR9 (named PcTLR9A and PcTLR9B) cDNA were cloned from the large yellow croaker, Pseudosciaena crocea. The full-length cDNA of PcTLR9A was of 3637bp, including a 5'-terminal untranslated region (UTR) of 111bp, 3'-terminal UTR of 355bp and an open reading frame (ORF) of 3171bp encoding a polypeptide of 1056 amino acids. However, the full-length cDNA of PcTLR9B was 119bp longer than that of PcTLR9A from the position of 3079-3197bp, which encoded a polypeptide of 1006 amino acids. Both of the PcTLR9A and PcTLR9B contained 12 typical structures of leucine-rich repeats (LRRs), an LRRTYP and an LRRCT in the extracellular region and a conservative Toll/IL-1R (TIR) domain in the intracellular region. However, compared to PcTLR9A, conservative Box 3 was absent in PcTLR9B TIR domain. Quantitative real-time reverse transcription PCR analysis revealed a broad expression of PcTLR9A and PcTLR9B with the highest expression in spleen and the weakest expression in muscle. Related Articles Rapid and reliable universal Cloning of influenza A virus genes by target-primed plasmid amplification. Nucleic Acids Res. 2008 Oct 2; Authors: Stech J, Stech O, Herwig A, Altmeppen H, Hundt J, Gohrbandt S, Kreibich A, Weber S, Klenk HD, Mettenleiter TC Reverse genetics has become pivotal in influenza virus research relying on rapid generation of tailored recombinant influenza viruses. They are rescued from transfected plasmids encoding the eight influenza virus gene segments, which have been cloned using restriction endonucleases and DNA ligation. However, suitable restriction cleavage sites often are not available. Here, we describe a Cloning method universal for any influenza A virus strain which is independent of restriction sites. It is based on target-primed plasmid amplification in which the insert provides two megaprimers and contains termini homologous to plasmid regions adjacent to the insertion site. For improved efficiency, a Cloning vector was designed containing the negative selection marker ccdB flanked by the highly conserved influenza A virus gene termini. Using this method, we generated complete sets of functional gene segments from seven influenza A strains and three haemagglutinin genes from different serotypes amounting to 59 cloned influenza genes. These results demonstrate that this approach allows rapid and reliable Cloning of any segment from any influenza A strain without any information abou ... Source: www.dailymail.co.uk --- 4 days ago

I think it's a MAJOR pain not having a "clone" feature in VMWare Server without using Virtual Infrastructure, so I made my own Linux shell scripts to do it. The clone script does a straight clone of a VM. It copies the directory, renames the files and tweaks all the text inside the files by using sed to replace the old name with the new one, then sets all the permissions to what they would be if you created a new VM. The rename script basically does all but copy it. This is handy for if you tried to rename a directory manually or attempted a manual copy and then found it still shows up under the old name in the web console. Remember this one assumes you have already renamed the directory to the new name and it MUST be the same as what you intend for the new name to be. Both work the same way... scriptname <old name> <new name> You can use quotes around the names if there are spaces, I went to great lengths to make sure it would still work with spaces in names (although I personally hate spaces). Save the scripts to the base directory where your VM's are, "standard" location is "/var/lib/vmware/Virtual Machines". Make executable with chmod +x <scriptname>. Once ran, add the new/renamed VM to the web console and choose "I copied it" or "I moved it" as appropriate. These scripts are offered free to everyone to use, modify, or do anything else you want with except take credit for the original version. Absolutely no warranty or guarantee o ... Australia issues Cloning license Australia has issued its first license for making stem cells from cloned human embryos, according to news reports. The recipient is a fertility company called Sydney IVF, which says it has access to 7,200 human eggs. The project will use only eggs obtained with appropriate informed consent and that could not be used for in vitro fertilization. Several research groups have already reported Cloning human embryos. However, despite multiple continuing attempts, no one has reported a successful dderivation of human embryonic stem cells through this method. The advantage of cloned hES cells would be that they would be genetically matched to an existing individual. The Cloning process could also help answer questions about `reprogramming’, or how cells shift genetic expression to become one or another type of cell. Recently, push to clone hES cells has diminished with the invention of a reprogramming technique that genetically engineers adult skin cells, but scientists are still eager to compare both techniques. See a blog entry describing previous attempts, plus links to other groups attempting this work in Europe. ... Source: www.associatedcontent.com --- 26 days ago

I need some advice for Cloning the hard drive of a G4 iMac to an external FW drive for temporary storage, in order to eventually clone the stored contents on to another G4 iMac. For more than one reason, a direct transfer between the two macs is not likely. There are some time constraints as the first iMac will wiped clean and given to a family member, while the "receiving" iMac is currently in pieces for upgrades/modding... which will probably not be finished before giving away the first iMac. FWIW, a dead monitor was removed and it worked fine. The monitor arm will be replaced with a handle, it will receive a new optical drive, larger HD, more ram, new battery, and a better fan... and maybe a glossy black paint job I just want to transfer everything, start up the "headless" iMac, and go about using it as though it were the original machine... without any "issues"... like having to re-install/activate software, passwords, users, etc., etc.. I'm looking for recommendations from those with experience with two or more programs, on the best software for doing this. Free or paid... just so long as it works without a hitch. And something that doesn't resorting to using the terminal. Many thanks in advance. ... Source: www.hackaday.com --- 6 days ago
