We have characterized the dynamics of beta-amyloid (Abeta) generation and deposition following gamma-secretase inhibition with compound LY-411575. Kinetic studies in pre-plaque mice distinguished a detergent soluble Abeta pool in brain with rapid turnover (half-lives for Abeta40 and Abeta42 were 0.7 and 1.7 h) and a much more stable, less soluble pool. Abeta in cerebrospinal fluid (CSF) reflected the changes in the soluble brain Abeta pool, while plasma Abeta turned over more rapidly. In brain APP C-terminal fragments (CTF) accumulated differentially. The half-lives for gamma-secretase degradation were estimated as 0.4 and 0.1 h for C99 and C83, respectively. Three different APP transgenic lines responded very similarly to gamma-secretase inhibition regardless of the familial Alzheimer's disease mutations in APP. Here, we report that the citrus bioflavonoid luteolin reduces amyloid-beta peptide (Abeta) generation in both Human "Swedish" mutant APP transgene-bearing neuron-like cells and primary neurons. We also find that luteolin induces changes consistent with GSK-3 inhibition that 1) decrease amyloidogenic gamma-secretase APP processing, and 2) promote presenilin-1 (PS1) carboxyl-terminal fragment (CTF) phosphorylation. Importantly, we find GSK-3alpha activity is essential for both PS1 CTF phosphorylation and PS1-APP interaction. I see Ontario Premier [Dalton] McGuinty is spending taxpayers dollars wisely once again in his province (from CTF blog ): Ontario Taxpayers Fund Sex-Change Operations Ontario began covering the bill for “sex-change” operations in 1971, but ceased doing so in 1998 under the Progressive Conservative government. In 2006, the Ontario Human Rights Commission required compensation for three patients midway through preparation for sex-change surgery during the 1998 insurance plan change. Now public funding is back regardless, at about $17,000 an operation. Health Minister George Smitherman estimates only ten Ontarians a year will have the surgery. I guess if there is one thing to be pleased with here, it is that this isn't the case in New Brunswick. Plus, it looks like one federal politician intends to stop federal cash from funding such procedures in the future. Glad to see someone has their head screwed on right.
