Scand J Immunol. 2008 Jul 21; Authors: Farkas L, Stoelcker B, Jentsch N, Heitzer S, Pfeifer M, Schulz C Chronic inflammation and acute exacerbations are pathophysiological features of chronic obstructive pulmonary disease (COPD). An impaired immune response to bacterial pathogens can contribute to both of them. Nucleotide oligomerization domain 2 (NOD2) is an intracellular receptor of innate immunity for muramyldipeptide (MDP). Mutations of the NOD2 gene followed by decreased recognition of MDP are associated with chronic intestinal inflammation and pulmonary complications of patients with allogenic stem cell transplant and sepsis. Our study provides evidence that NOD2, toll-like receptor 4 (TLR4) and the adapter protein receptor-interacting protein 2 (RIP2) are induced by tumor-necrosis factor-alpha (TNF-alpha) and interferon-gamma (IFN-gamma) in the bronchial epithelial cell line Beas-2B. We also demonstrate that lipopolysaccharide (LPS) can further increase NOD2 transcription in a TNF-alpha and IFN-gamma-induced activation state. In addition, we show that, while MDP fails to enhance CXCL-8 release from otherwise unstimulated Beas-2B cells, a 12 h prestimulation period with TNF-alpha and IFN-gamma primes the cells for an additional increase of CXCL-8 secretion via induction of NOD2 and RIP2. Went to Beas River for the Olympics today, which was great fun. Anyway, as we have never been at an equestrian event, we (and many others) brought an umbrella along, which became handy as it was drizzling on and off. Simple ponchos were actually provided at the venue, but we felt that with the rain being inconsistent, and the poncho creating an unbearable green house effect, it would be easier to use the umbrella when necessary. The venue was huge, visitors were spread out nicely over the entire course, and we made sure not to block the view of other spectators when we used the umbrella. This worked well for about 30 minutes, and many other people did the same. After standing for a while at one quiet spot, one of the volunteers suddenly asked us not to use the umbrella. As he walked away immediately, we were not able to ask for a reason but complied, and thought we were probably blocking somebody's view unknowingly. 15 minutes later we were walking to another part of the venue, again with umbrella. This time a seemingly more senior western official demanded rather rudely that we not use the umbrella. I asked him why, and he said it is not allowed as it would scare the horses. I pointing out to him that many people were using it and that I was not aware of this rule, but he was not impressed. Well, we made it Manali. The bus journey was rubbish, other than a couple of hours of sleeping strapped to the boats on the roof. Turns out this is pretty illegal, so it’s a good thing we weren’t noticed! We’ve been based out of a lovely hotel, hot showers, cider, comfy beds. It’s been lovely. I’m currently stuffed from a curry and Baskin Robins icecream. Oh and a bloke gave a beggar a good kicking. What a super place! But I digress…. When Tim arrived, several of us jumped on a few km of the Manalsu behind Old Manali. Shrinkavision kicking in nicely it proved to be pretty blind, super continous and really good fun. A few chunky pourovers to get beaten in, and plenty of tasty lines. Just the job before some cider and momos! (yes Liz, they are ace!) Today we ran a few sections of the Beas. Patrick and Tom H were feeling a little worse for wear and so decided to get on lower. It seems they had the best time… The rest of us put on a swollen grey river that was thundering along at a fair old rate. After a km of so we reached a (more) blind rapid and pulled in for a scout. It looked like a nice chunky rapid, a few bigger holes to dodge but nothing major, before (appearing to) flatten off and provide a big eddy. We all dropped in and made it through to the corner, but it didn’t flatten off. Hong Kong The most serious concern here, as in Beijing, is the weather. The skies are blue but come 11 o'clock in the morning, the heat is so oppressive that half an hour outside is as much as anyone not accustomed to it can bear. The cross-country section of the three-day-event on Monday begins early in the morning but is unlikely to have finished until 11.30am so the later starters will be at a serious disadvantage, unless the Beas River location is much cooler than here at Sha Tin . At a distance of only 5.7km it is already the shortest cross-country course in Olympic memory and discussions are taking place about the possibility of it being shortened further. Having said that, conditions can change very quickly and with rain forecast for the next five days and talk of a typhoon at the weekend , the stamina-sapping heat may be the least of the problems. On the plus side, the facilities are top class, funded by the Hong Kong Jockey Club at a cost of £53m, and all of the horses have travelled over well. The grooms on the flight with them reported that they had never known a smoother journey and that the transfer of the horses into air-conditioned horseboxes and then to the stables took only an hour. The horses have been chosen not just for ability, they are all capable of a top ten performance, but also for their build. Today I was able to check out the cross country course located at Hong Kong's Beas River Golf Course just a few miles from the old border with China. I've been really looking forward to it as the cross-country phase is my favourite part of a three-day event. I love the atmosphere on the course, the huge, solid obstacles and the speed and bravery required of the riders. Before I got to see the course I caught up with Team GB's eventers who had just walked it themselves. Mary King told me it was ''difficult'' and when I said: ''It'll sort out the men from the boys then?'' She answered: ''Yes and let's hope we're the men!'' My first impressions of the course were that it's very small. The grassy course weaves and loops across fairways and round bunkers but it's not the vast open space you get at Badminton or Burghley . We were lucky enough to be part of a media tour with the course designer Mike Etherington-Smith. It was really interesting and yes, we did actually walk it, although not quite the full 5.7km. Luckily the weather was kind to us, not too hot and no rain which was lucky as I have lost my waterproof. Mike told me the actual track the horses will gallop on cost hundreds of thousands of pounds because they have put in drainage throughout the whole course.
