Background: Aneuploidy is a feature of most cancer cells that is often accompanied by an elevated rate of chromosome mis-segregation termed chromosome instability (CIN). While CIN can act as a driver of cancer genome evolution and tumor progression, recent findings point to the existence of a threshold level beyond which CIN becomes a barrier to tumor growth and therefore can be exploited therapeutically. Drugs known to increase CIN beyond the therapeutic threshold are currently few in number, and the clinical promise of targeting the CIN phenotype warrants new screening efforts. However, none of the existing methods, including the in vitro micronuclei (MNi) assay, developed to quantify CIN, is entirely satisfactory. Methods: We have developed a new assay for measuring CIN. This quantitative assay for chromosome mis-segregation is based on the use of a non-essential human artificial chromosome (HAC) carrying a constitutively expressed EGFP transgene. Thus, cells that inherit the HAC display green fluorescence, while cells lacking the HAC do not. This allows the measurement of HAC loss rate by routine flow cytometry. Results: Using the HAC-based chromosome loss assay, we have analyzed several well-known anti-mitotic, spindle-targeting compounds, all of which have been reported to induce micronuclei formation and chromosome loss. I remember the first time I prepared to go to Haiti -- I told an acquaintance about the trip and they responded with a sincerely puzzled, "why are you going there?" I didn't have a good answer other than that it felt right to take action around the events of the 2010 earthquake that killed over 220,000 people . I had the skills to mobilize people. I could secure needed funds and generate support for community organizations in-country. I also had the backing of a travel organization committed to developing an effective volunteer program . I wondered if there would be interest. Who would be willing to give up the "idyllic beach vacation" or "adventure travel tour" and volunteer their time and energy to help others in one of the poorest countries in the world? I was surprised and heartened to receive hundreds of requests from people all over the world wanting to give their time and money to help. Chicago Bears general manager Phil Emery had his work cut out for him this offseason. He had to upgrade the offensive line, get more weapons for quarterback Jay Cutler and then find a coach who could improve Cutler's play. On paper, it appears that he's accomplished all three goals, but the question is, how will it all work out when the pads are strapped on? It's no secret that the Bears need Cutler to step his game up. Ever since he was traded to the Windy City in 2009, he's been viewed as the franchise quarterback, a position the team hasn't had much success at. With Cutler, they received a passer with unreal arm strength, uncanny ability to throw from various platforms and one of the league's toughest players. They also received one of the league's most frustrating players, however, and arguably one of its most inconsistent quarterbacks. Cutler threw only 19 touchdowns and 14 costly interceptions in 15 games last season. The latter number isn't high, but many of the turnovers came at the wrong time. Part of the reason is because of his decision making and mechanics. He tends to go flat-footed in the pocket, consequently taking more time to rotate his hips—when he actually does it—and transfer his weight. The U.S. Coast Guard Academy Division of Athletics announced today that it will elevate the men’s and women’s club lacrosse teams to varsity status, effective with the 2014-15 academic year. The teams will compete officially for the first time as Division III intercollegiate squads playing in the NEWMAC Conference in the spring of 2015. “This is a very significant stride for the CGA athletics program,” said Director of Athletics Tim Fitzpatrick. “It’s a great thing to be able to accommodate the interests and talents of so many cadets by bringing two very strong club programs to the varsity level. The popularity and growth of lacrosse both regionally and nationally in the recent past is very well documented. We look forward to the addition of the two lacrosse programs helping us attract more of our nation’s best and brightest student-athletes to the academy.” The men’s lacrosse club was started by John Senyard (USCGA, Class of 1983), and competed for the first time in the spring of 1984. I understand what you’re saying, to a certain extent. I love the entire BRF, and while I may personally feel more inclined to a few members, I still understand there is a hierarchy not defined by people’s opinions, but by status, which is important to adhere to.
